| Testing Indicators | Appearance — Visual inspection (white to off-white crystalline powder); Melting point — Capillary method (208–213°C, literature; 221.0–225.0°C, TCI specification); Purity — HPLC-UV (≥96–99.5%, C18 column, acetonitrile:buffer gradient, λ=220–254 nm); Related substances — HPLC (impurity profiling including venlafaxine, O-desmethylvenlafaxine related compounds A/B/C, and other degradation products); Assay — RP-HPLC or nonaqueous titration (TCI: min. 96.0% by nonaqueous titration; USP: 90.0–110.0% of label claim for extended-release tablets); Loss on drying — Gravimetric (≤0.5%); Residue on ignition — Gravimetric (≤0.1%); Heavy metals — Colorimetric/ICP-MS (≤20 ppm); Identification — FTIR (characteristic peaks: O–H (phenol) ~3300–3500 cm⁻¹, O–H (alcohol) ~3200–3400 cm⁻¹, aromatic C–H ~3000–3100 cm⁻¹, aliphatic C–H ~2850–2950 cm⁻¹, aromatic ring ~1600 cm⁻¹, C–O ~1050–1250 cm⁻¹); UV spectrophotometry (pH-dependent solubility); ¹H NMR (DMSO-d₆ or CDCl₃: δ 2.20 ppm singlet N(CH₃)₂, δ 2.60–2.80 ppm multiplet CH₂N, δ 3.20 ppm multiplet CH(cyclohexyl), δ 6.60–7.10 ppm multiplet aromatic protons, δ 9.20 ppm broad singlet phenolic OH); ¹³C NMR (DMSO-d₆: δ 40 ppm N(CH₃)₂, δ 55 ppm CH₂N, δ 70 ppm C–OH (cyclohexyl), δ 115–130 ppm aromatic carbons, δ 155 ppm C–OH (phenol)); Mass spectrometry — ESI-MS (m/z 264 [M+H]⁺); Water content — Karl Fischer titration (GB/T 6283/EP 2.5.12); pH — Potentiometry (aqueous suspension, weakly basic due to tertiary amine, pH-dependent solubility); Specific optical rotation — Not applicable (racemic mixture, RS configuration); Dissolution — USP apparatus (for extended-release tablets, pH 6.8 phosphate buffer, 75 rpm, 900 mL). |
| Storage Notes | Store in a cool, dry, well-ventilated area at 2–8°C. Protect from light, moisture, and air. Keep container tightly sealed. Store powder at -20°C for long-term stability (up to 3 years); at 4°C for 2 years. In solvent, store at -80°C for 6 months or at -20°C for 1 month. Light-sensitive and hygroscopic material; avoid prolonged exposure to light and humid conditions. The solubility of desvenlafaxine is pH-dependent; store away from strongly acidic or basic conditions. Store away from strong oxidizing agents and incompatible substances. |
| Lead Time | 10 days |
Desvenlafaxine (INN, also known as O-desmethylvenlafaxine, ODV, or WY-45233) is the major active metabolite of the antidepressant venlafaxine and a structurally novel serotonin-norepinephrine reuptake inhibitor (SNRI). It functions as a potent and selective inhibitor of neuronal serotonin (5-HT) and norepinephrine (NE) reuptake, with Ki values of 40.2 nM and 558.4 nM, respectively, and weak inhibition of dopamine reuptake. Used as a pharmaceutical intermediate and active pharmaceutical ingredient (API) for the development and quality control of extended-release tablet formulations (e.g., Pristiq®, Khedezla®). Approved for the treatment of major depressive disorder (MDD) in adults. Also employed in analytical method development, method validation (AMV), forced degradation studies, and abbreviated new drug application (ANDA) support. ATC code: N06AX23.
